中等强度运动训练结合仙灵骨葆胶囊对去卵巢大鼠骨密度、骨代谢及生物力学的影响

    摘要 目的:以去卵巢大鼠为研究对象,观察中强度运动训练结合仙灵骨葆胶囊对去卵巢大鼠骨密度、骨代谢及股骨生物力学的影响。方法:50只雌性SD大鼠按照随机数字表法分为5组,A组为正常对照,B组切除卵巢1周后灌胃给予生理盐水1mL,C组切除卵巢1周后进行中等强度的运动训练(运动速度为20m/min,每天持续时间为60min,连续5d/周),D组切除卵巢1周后灌胃给予仙灵骨葆胶囊[0.4g/(kg?d)]1mL,E组切除卵巢1周后灌胃给予仙灵骨葆胶囊1mL+中等强度的运动训练,每组10只。连续治疗8周后,进行血液生化指标检测,以及股骨、L5椎体的骨密度、Micro-CT检测与生物力学检测。结果:与B组比较,C~E组的BALP、PINP、骨钙素浓度明显升高(P<0.05),血钙、β-CTx、TRACP-5b浓度明显降低(P<0.05);与C、D组比较,E组的BALP、PINP、骨钙素浓度明显升高(P<0.05),血钙、β-CTx、TRACP-5b浓度明显降低(P<0.05)。与B组比较,C~E组的L5椎体、股骨骨密度明显升高(P<0.05);与C、D组比较,E组的L5椎体、股骨骨密度明显升高(P<0.05)。与B组比较,C~E组股骨(或L5椎体)的骨体积分数、骨小梁数目、骨小梁厚度明显升高(P<0.05),骨小梁间隙与结构模型指数明显降低(P<0.05);与C、D组比较,E组股骨(或L5椎体)的骨体积分数、骨小梁数目、骨小梁厚度明显升高(P<0.05),骨小梁间隙与结构模型指数明显降低(P<0.05)。与B组比较,C~E组L5椎体的最大载荷、最大挠度及最大应力明显升高(P<0.05);与C、D组比较,E组L5椎体的最大载荷、最大挠度及最大应力明显升高(P<0.05)。与B组比较,C~E组股骨的最大载荷、弹性载荷、弹性挠度、弹性模量、弹性应力、最大应力及弹性挠度显著升高(P<0.05);与C、D组比较,E组股骨的最大载荷、弹性载荷、弹性挠度、弹性模量、弹性应力、最大应力及弹性挠度显著升高(P<0.05)。结论:中等强度运动训练结合仙灵骨葆胶囊治疗可提升去卵巢大鼠的骨密度,改善骨代谢与骨微结构,提高骨力学性能,二者具有协同作用。

    Abstract:

    Objective:Taking ovariectomized rats as the research object, the effects of moderate intensity exercise training combined with Xianlinggubao Capsule on bone mineral density, bone metabolism and femoral biomechanics of ovariectomized rats were observed.Methods:Fifty female SD rats were randomly divided into 5 groups: group A was the normal control; group B was given 1ml normal saline by gavage after ovariectomy for one week; group C was given moderate intensity exercise training (exercise speed was 20m / min, lasting for 60min per day, continuous 5D / week); group D was given Xianlinggubao capsule [0.4g / (kg · d)] 1ml after ovariectomy, and group E was ovariectomized for 1 week Four weeks later, Xianlinggubao capsule (1ml) and moderate intensity exercise training were given by gavage, with 10 rats in each group. After 8 weeks of continuous treatment, blood biochemical indexes, bone mineral density (BMD), micro CT and biomechanics of femur and L5 vertebral body were detected.Results:Compared with group B, the concentrations of BALP, PINP and osteocalcin in group C ~ e were significantly increased (P < 0.05), and the concentrations of serum calcium, β - CTX and tracp-5b were significantly decreased (P < 0.05); compared with group C and D, the concentrations of BALP, PINP and osteocalcin in Group E were significantly increased (P < 0.05), and the concentrations of serum calcium, β - CTX and tracp-5b were significantly decreased (P < 0.05). Compared with group B, the BMD of L5 vertebral body and femur in group C ~ E was significantly increased (P < 0.05); compared with group C and D, the BMD of L5 vertebral body and femur in Group E was significantly increased (P < 0.05). Compared with group B, the bone volume fraction, trabecular number and trabecular thickness of femur (or L5 vertebral body) in group C ~ E were significantly increased (P < 0.05), and the trabecular space and structural model index were significantly decreased (P < 0.05); compared with group C and D, the bone volume fraction, trabecular number and trabecular thickness of femur (or L5 vertebral body) in Group E were significantly increased (P < 0.05), and the trabecular space and structural model index were significantly decreased (P < 0.05) .05)。 Compared with group B, the maximum load, maximum deflection and maximum stress of L5 vertebral body in group C ~ E were significantly increased (P < 0.05); compared with group C and D, the maximum load, maximum deflection and maximum stress of L5 vertebral body in Group E were significantly increased (P < 0.05). Compared with group B, the maximum load, elastic load, elastic deflection, elastic modulus, elastic stress, maximum stress and elastic deflection of femur in group C ~ E were significantly increased (P < 0.05); compared with group C and D, the maximum load, elastic load, elastic deflection, elastic modulus, elastic stress, maximum stress and elastic deflection of femur in Group E were significantly increased (P < 0.05).Conclusion:Moderate intensity exercise training combined with Xianlinggubao capsule can improve bone mineral density, bone metabolism and bone microstructure, and improve bone mechanical properties of ovariectomized rats.
