液态介质环境对乳腺癌细胞弹性模量的影响

    乳腺癌的新发病率目前位居女性恶性肿瘤发病率之首,急需研发新的诊疗方法,体外细胞实验是研发中关键的一环。体外细胞实验中需要采用液态培养介质,常用的三种介质为PBS缓冲液、DMEM培养液和DMEM+10%FBS培养液。然而,液态介质环境对于乳腺癌细胞力学性能的影响还未见文献报道。本文通过原子力显微镜技术测试了乳腺癌细胞(MCF7)在上述三种液态介质环境中的弹性模量,分别为2586.91±663.45 Pa、2106.86±692.07 Pa、1586.52±794.01 Pa,存在显著差异(p<0.001)。其中PBS缓冲液中的弹性模量是DMEM+10%FBS培养液中的1.63倍,这个差异甚至接近同一种液态介质环境中健康乳腺细胞和发生癌变的乳腺细胞的差异。同时测量了乳腺癌细胞在三种液态介质环境中的细胞粘附力,分别为63.81±24.50 pN、66.09±26.41 pN、121.97±43.99 pN,在DMEM+10%FBS培养液中的粘附力与另外两种液态介质存在显著差异 (p<0.001)。

    Abstract:

    The new incidence of breast cancer ranks the first in the incidence of female malignant tumors. It is urgent to develop new diagnosis and treatment methods. In vitro cell experiments, liquid media were needed, and the three commonly used media are PBS, DMEM culture medium and DMEM+10% Fetal Bovine Serum (FBS) culture medium. However, the effect of liquid medium environment on the mechanical properties of breast cancer cells is rarely reported. Using atomic force microscope (AFM), we measured the elastic modulus of breast cancer cells in the above three kinds of liquid medium environment. The elastic modulus are 2586.91±663.45 Pa, 2106.86±692.07 Pa and 1586.52±794.01 Pa. The elastic modulus in PBS is 1.63 folds of that in DMEM+10%FBS, a difference close to that of healthy and cancerous breast cells in the same liquid medium. The cell adhesion force to AFM probes in the above three liquid media environments are 63.81±24.50 pN, 66.09±26.41 pN and 121.97±43.99 pN, respectively. Cell adhesion force in DMEM+10%FBS medium is significantly different from that of the other two liquid media.
